Identifikasi dan isolasi promoter gen pembungaan kakao TcLFY Identification and isolation for promoter of TcLFY cacao flowering gene

Summary
Promoter is a regulator of gene
expression for a phenotype or trait carried by
the gene. In the structure, a promoter located
beyond the 5’ end of the open reading frame of
the gene on which its expression is regulated.
This research aimed to isolate the DNA
fragment flanking TcLFY at the 5’ end and to
analyze whether the fragment has charac-
teristics of the promoter, primarily the core
motifs of promoter. Using Genome Walking
technique, DNA fragments flanking the TcLFY
gene at its 5’ end was isolated. Analysis of the
DNA sequence was done using online
computer software accessible through web site
www.softberry.com and an entry sequence of
the flanking DNA fragment along with the 2.5
kb TcLFY sequence. The result indicated that
the flanking fragment has core motifs for a
promoter at proper positions, which are TATA
box at position –80, CAT boxes (CCAAT) at -
387 and –626, and GC boxes that are known as
UAS were found at the -323 and –537
positions. To obtain a conclusive result, this
promoter sequence needs to be further
examined to confirm its function.
Ringkasan
Promoter merupakan pengendali ekspresi
gen untuk memunculkan fenotipe atau karakter
yang dibawa oleh gen tersebut. Di dalam
strukturnya, promoter umumnya terletak di
daerah ujung 5’ gen yang dikendalikan
ekspresinya. Tujuan penelitian ini adalah
mendapatkan fragmen DNA yang mengapit
gen pengendali pembungaan kakao (TcLFY)
dan menganalisisnya apakah memiliki
karakteristik promoter, yaitu mengandung
motif-motif inti (core motifs) dari promoter.
Dengan teknik Genome Walking, fragmen
DNA pengapit gen TcLFY di ujung 5’ dapat
diisolasi. Analisis sekuen menggunakan
perangkat lunak komputer online (www.
softberry.com) dengan input data fragmen
tersebut ditambah gen TcLFY 2,5 kb di
bawahnya, mengindikasikan adanya beberapa
motif inti promoter pada posisi yang sesuai,
yaitu kotak TATA pada lokasi –80, kotak CAT
(CCAAT) di posisi -387 dan –626, dan kotak
GC yang merupakan UAS dijumpai pada
lokasi -323 dan –537. Untuk memperoleh hasil
yang bersifat konklusif, sekuen promoter ini
masih perlu diuji fungsinya.
