Abstract
The utilization of latex effluent to produce
bioproduct like indole acetic acid (IAA) will
reduce amount of effluent, as well as effluent
processing cost and produce an economically
profitable product. IAA could be produced by
some rhizosphere microbes that could grow on
latex effluent using L-tryptophan (Trp) as its
precursor. The aim of this research is to
determine potential growth and capability of IAA
production by Rhizobium spp. R6 and KT on
synthetic and latex serum media supplemented
with pure Trp and with litter poultry manure as a
cheap source of Trp. The research covered
examination of IAA producing Rhizobia using
liquid synthetic media supplemented with 0.07
g/L and 0.14 g/L Trp. The potential Rhizobium
sp. in producing IAA was then inoculated into
latex serum media supplemented with pure Trp
and Trp from litter poultry manure. Result of the
research showed that the highest IAA production
was reached as much as 51.08 µg/mL in synthetic
media supplemented with 0.14 g/L Trp inoculated
with Rhizobium sp. R6. IAA could be produced
as much as 6.63 µg/mL in pasteurized
undiluted latex serum media supplemented with
0.14 g/L Trp. Using latex serum media
supplemented with Trp from litter poultry manure
showed that Rhizobium sp. R6 could produce
11.91 µg/mL. Supplementation of pure synthetic
Trp in IAA production could be replaced with
litter poultry manure as a cheap source of Trp.
Abstrak
Pemanfaatan limbah lateks menjadi produk
bio seperti asam indol asetat (IAA), dapat
mengurangi volume limbah, menekan biaya
pengolahan limbah, serta menghasilkan produk
yang bernilai ekonomis. IAA dapat dihasilkan
oleh beberapa mikroba rhizosfer yang mampu
tumbuh dalam limbah lateks dengan L-triptofan
(Trp) sebagai prekursor-nya. Penelitian bertujuan
menetapkan potensi pertumbuhan dan produksi
IAA oleh Rhizobium spp. R6 dan KT dalam
medium sintetik dan serum lateks yang
disuplementasi Trp sintetik dan kotoran ayam
sebagai sumber Trp murah. Isolat potensial dalam
produksi IAA kemudian ditumbuhkan dalam
medium serum lateks pekat yang disuplementasi
Trp murni dan Trp dari kotoran ayam. Hasil
penelitian menunjukkan bahwa produksi IAA
tertinggi diperoleh dalam medium sintetik oleh
Rhizobium sp. R6, sebesar 51,08 µg/mL. IAA
dapat diproduksi sebesar 6,63 µg/mL dalam
medium serum lateks 100% + Trp 0,14 g/L yang
dipasteurisasi. Dalam medium serum lateks yang
disuplementasi Trp dari kotoran ayam, produksi
IAA Rhizobium sp. R6 dapat mencapai 11,91
µg/mL. Suplementasi Trp murni dalam produksi
IAA dapat digantikan dengan kotoran ayam
sebagai sumber Trp yang mura
